It is possible Petey has built up a tolerance to Trazodone. This can happen if used long term. Discuss an increase with your veterinarian. Consider also desensitization therapy and a pheromone product, such as the Adaptil collar or DAP plug in diffuser, to further help decrease anxiety.

How about trying something like Trazodone instead? Trazodone works to increase Seratonin in the brain, and can cause relaxation and even mild sedation at high doses. I prefer to use this for cases of anxiety. If you choose to persist with Melatonin, I would use a reputable company like Thorne.

There should be no harmful interactions at all between tryptophan and chamomile, and the prescription medications that your dog is currently taking - so you could certainly try this if you like. I do recommend letting your vet know that you are giving the supplement, just so that he/she is in the loop about everything your dog is taking :)

Yes, trazodone is often given daily to help with anxiety. There are many options however. The most common medication for separation anxiety is Fluoxetine. It's generally my first choice when I'm looking for a daily medication. I recommend to discuss this with your veterinarian. Also, medication by it's self is not likely to work. Exercise and behavioral modification are also needed. Hyperactivity can be a side effect of Trazodone. Here is an information sheet about this drug and list of the side effects. Unfortunately your dog may be one of the ones that does not react as expected to this drug. You will need to speak to your vet about possibly trying something different.
