The most likely side effect would be vomiting or GI upset, in the first day or two after taking the meds. If she's acting normal so far, you probably won't see any issues at all. It's fine for her to continue with her normal daily routine as far as eating, exercising, etc.

Diarrhea can be a side effect of any antiparasitic medication. As long as Butter is behaving normally otherwise, you can manage it at home. Withhold food for 12-24 hours. Allow small amounts of water or unflavored PediaLyte. Resume feeding a bland diet in small, frequent amount until symptoms resolve. Begin with 1 tablespoon of food every hour and gradually increase the amount. Feed bland until the stool is normal then transition slowly to the regular diet. Symptoms to monitor for and that should be addressed by your veterinarian include lethargy, vomiting, persistent diarrhea, loss of appetite or abdominal pain.

If there are no other symptoms and she is still eating and drinking normally you can probably wait until Monday to have her examined. I doubt the recent vaccines and Trifexis are related. If she vomits or has diarrhea, if she isn’t eating or if she is so lethargic you can’t rouse her you should see an emergency vet.
