Dog has red spot with hole after scab came off. What could it be?

This could be a skin infection caused by a fungus or bacteria or and old injury or bite and in worst case scenario it could be a tumor. Please have this examined by your veterinarian. If he suspects a tumor, he will likely want to remove it and send it in to determine whether it is malignant and your dog needs further treatment. If it is an infection your dog may need an antifungal or antibiotic ointment. It may just be a scab from an old wound and might need no treatment. Your vet will be able to determine that with a hands-on exam.

Poor guy. This looks like a "hot spot" or superficial pyoderma (infected area) on the skin. This can be caused by an initial knick or scrap on the skin that later got infected and scabbed over. Flea bites are a common cause of hot spots, so its best to keep Bono on flea prevention year round it they are common in your area. Your vet should be able to prescribe some oral antibiotics to help this resolve completely. The wound spray you have may be sufficient, so its best to take it in with you to Bono's vet appt for your vet to see. If not, then the vet will likely recommend another topical to use. However, other things are possible. I recommend that you have Candy evaluated by your veterinarian for further assessment and treatment. I recommend having Roscoe seen by a Vet, as cysts can become infected when they rupture, and Roscoe may need a course of antibiotics. Your vet can then let you know if they have any concerns about the lesion when they’re able to examine it. In the meantime, keep it clean using soapy water (Dawn dish soap works well) and soak/wipe it clean a few times daily. Hope this helps!

The wound looks puffy from the photo (but I would need to palpate it to know for sure). The crusty area next to it could indicate another wound that is scabbed over or it could indicate a bacterial infection or ringworm (which humans can get). Your best option is to have Sophie examined by the veterinarian in case the wound is infected. She would be placed on antibiotics if that is the case. They could also test her for ringworm if it looks suspicious on exam. Make sure to protect the wound from scratching or licking. Sophie may need to wear an elizabethan collar if she can lick the area. A doggie shirt can help protect it from scratching. You do not want to disrupt the scab (it's nature's bandaide).
