Should I separate my male kitten from his mom during and after birth?

Pet's info: Cat | Mixed Breed | Female | unspayed | 15 lbs

My Lola stink is pregnant with her 2nd litter I kept a male kitten from her first litter he (puzzle) is now 4 months old . When Lola does go into labor will I have to keep puzzle away from Lola and her kittens ?

Yes, you will need to Puzzle away, and I would keep them complete separated until the kittens are older and starting to wean. I would also neuter Puzzle so he doesn't mate with his mother. Best of luck.
