Is my 3-month-old Persian kitten single or double coated? What's on its nose?

It looks like he may have a dermatitis on his nose which could be secondary to a bacterial infection, trauma, allergic reaction, or possibly a fungal infection. He should be examined by a vet to determine the cause. Your vet can do a culture, if needed to help diagnose.

With this breed and the signs I would be worried about ringworm. Persian cats and other long-haired breeds are prone to this type of fungal infection and in fact the mother may have it and be what's called an asymptomatic carrier. I recommend practicing strict hygiene protocols with handling the kittens and discussing with your veterinarian if they feel they should be testing for ringworm. I agree with your veterinarian that this is unlikely to be a side effect of the medication previously given.

I see what you mean about the dark area on Piper Grace's nose. This could be due to something like ringworm or a skin infection. Without seeing her in person, I can't be certain what is going on, so I recommend a vet visit for her. The vet can diagnose what is causing this problem and can treat her. Persian kittens are very prone to ringworm, a fungal infection. I suggest you take Lily to your veterinarian for a skin exam. She is due for vaccinations anyway so show the skin lesion to your vet. Your vet will want to look at the lesion with a black light and/or do a brush test and try to grow the fungus as a diagnostic to determine if it is indeed a fungal infection. As long there is no definitive diagnosis, please make sure you wash your hands after petting your cat, as ringworm is contagious to people.

A picture of the skin would help a lot here to help classify what this likely is. With your description however, it sounds likely that this may be either a bacterial or fungal infection. Bacterial infections are often caused by a wound on the skin that didn't heal properly. I would have your vet take a look at the area or repost with an actual picture so that we can give you more advice on treatment. This could be due to a localized infection. One possibility (considering the recent illness) is a form of calicivirus that attacks the skin. This usually resolves on its own within 2-3 weeks, but may be complicated by secondary infections. This could also be due to a bacterial or fungal infection. It's best to take Miso to the vet (possibly the shelter vet), so they can try to identify the cause of this. For now, you can help by keeping the nose clean. You can do this by gently wiping with a cloth and warm water twice a day. You can then apply a triple antibiotic cream to the affected area after each wash.
