If you are asking about vaccinations at the Vetco Vaccine clinics located in some Petco stores, you do not need an appointment, you just show up at the designated time and it is first come first served. You can look at the schedules near you at:

Other veterinary clinics may require an appointment and you will need to call them directly to find out.

You do not have to make an appointment, you can just show up. However since your puppy is so young, I strongly suggest you do not touch any other dogs, nor let anyone touch your puppy as there is always a possibility for your puppy to catch an infection, especially parvo. Bacteria and viruses can be transmitted by petting. Take her in, have her vaccinated, take her out and home.

Petco Pet Education Center, formerly Petcoach has two physical locations. These are normal clinics that see both healthy and sick pets. Petco offers vaccine clinics. These are for healthy dogs who just need routine care and are walk in style. I recommend to look at the Petco and Petco Pet Education Center, formerly Petcoach websites for clinic information.
