Prune juice will not harm your dog. If you are using it as a laxative, it isn't the best one to use. Try increasing water intake by soaking food in warm water or mixing in some water. Offering a 50/50 solution of water and unflavored PediaLyte may also help. See your vet if your dog begins straining to defecate, vomiting or loses his appetite.

It is not recommended and may actually cause gastric upset if given. If your dog is constipated and you want to increase the fiber in his diet you could add 1 tablespoon of canned pumpkin to his diet and see if that helps. Otherwise, your vet can prescribe medications to help with constipation.
