Great question! It's actually a relatively easy process. First thing to do is to place the litter box in a private area away from their food and water. Kittens do not like use the litter box next to their food and water. Use a litter box with a low lip so the kittens can easily step over it to get into. Place them in the litter box a couple of times, and scratch the litter with your finger. Do this immediately after they eat, and they will learn very quickly that the litter box is their toilet.

Kittens usually need to eliminate after feeding, so every time after feeding her and every time you think your kitten may want to go, gently put it in the litter box, gently pick up one of the front legs and scratch the litter with it. If and when your kitty then pees or poops, let it finish and then praise it . They usually learn very fast. It is usually their mother that teaches them when and how to use a litter box.
Your kitten probably was taken away from its mother too early.
