What's this lump on my 2-week-old puppy's leg, on nursing replacement?

This is often differentiated by a test called a fine needle aspirate (FNA) which is a pain free, inexpensive and easy procedure which in most cases will help reach a diagnosis. It's really difficult to know what this is without seeing it. It could be related to an infection, could be a fluid swelling from getting hurt, or a congenital abnormality. I doubt it's something contagious, but I can't know for sure without seeing it. If the pup is otherwise acting normal, then it's not an emergency and can wait until the next day to see you regular vet. No puppies don't get fatty lumps like this overnight, so it's more likely a cyst or an abscess on Franko. A cyst and an abscess are not serious, but I would still have it checked out by a vet before his next round of vaccines. The vet can do a fine needle aspirate on it. To do this, the vet sticks a small needle into the lump to get a sample of it's cells, and examines the cells on a slide under a microscope. It's usually painless to the animal, and relatively quick to perform. Once the cause of the lump is diagnosed, the vet can discuss treatment options.

It sounds like this wound that became a lump that is oozing fluid has become an abscess, or a pocket of infected fluid. Abscesses form when infection is trapped under the skin or within tissues such as fat or muscle. The bacteria will try to find a way out, so they can break through the skin and create a "draining tract" of pus. This should be addressed by your veterinarian, who will likely drain the abscess and open it up to allow continuous drainage.
