My 2 yr old Labrador is approx 27kg and has eaten approx a teaspoon of cooked onion. Is that enough to harm him?

Luckily for your pup, the amount of onion ingested is very unlikely to cause any major clinical consequences. For his size, he would need to eat at least two medium sized raw onions to experience toxicity. I would continue to monitor him and make sure he doesn't develop digestive upset from the soup.
